LSD is one of the most potent 5-HT2A agonists known. At sub-perceptual doses it nudges the same machinery that drives learning and plasticity — without the full perceptual experience.
Sublingual uptake
Held under the tongue, the compound absorbs through oral mucosa into the bloodstream and crosses the blood–brain barrier within minutes.
5-HT2A binding
LSD binds serotonin 5-HT2A receptors as a partial agonist (with activity at 5-HT2C, 5-HT1A and dopamine D2). Cryo-EM imaging shows the molecule sits unusually deep in the receptor — a structural "lid" that explains its long, slow action.
Cortical glutamate release
Activation of 5-HT2A on layer-V pyramidal neurons in the prefrontal cortex increases glutamatergic signalling — the opening move of the brain's growth response.
The plasticity cascade
Downstream BDNF → TrkB → mTOR signalling promotes dendritic branching and synapse formation. Classic psychedelics are "psychoplastogens" — they transiently raise the brain's capacity to rewire (Ly & Olson, 2018).
Sub-perceptual by design
At 5–10μg you should not feel "high." The intent is a quiet lift in mood, focus and flexibility — enhanced, not altered. If you can clearly tell you took something, you took too much.
NETWORK FLEXIBILITY
At higher doses LSD loosens the brain's default-mode network and increases cross-network communication. At microdoses these effects are far subtler, but the same flexibility is the proposed basis for shifts in creativity and perspective.
WHAT THE EVIDENCE SAYS
Open-label and survey data on microdosing are broadly positive for mood, focus and creativity. But placebo-controlled trials (e.g. Szigeti 2021; de Wit's lab) suggest a large share of the benefit is expectancy. Treat microdosing as promising and under-studied — not settled science.
Lysergic acid diethylamide — the lysergamide first synthesized by Albert Hofmann at Sandoz in 1938. Its tetracyclic ergoline scaffold locks deep into the serotonin 5-HT2A receptor, where an extracellular "lid" slows its release and gives LSD its long, characteristic action.

FADIMAN
Day 1 dose · Day 2 transition · Day 3 rest, then repeat. The most common protocol. Off-days let 5-HT2A receptors re-sensitise and help you read effects against a clean baseline.
STAMETS-STYLE
Four days on, three days off — adapted from the psilocybin "stack." Often paired with Lion's Mane and a low dose of niacin (see the stack below).
EVERY-THIRD-DAY
A simple fixed cadence (dose every third day) favoured by the Microdosing Institute. Whichever you choose, cycle 4–8 weeks then take a 2–4 week break to avoid tolerance and habituation.
"Stacking" pairs a microdose with supportive compounds thought to extend neuroplasticity and smooth the experience. Evidence is early — treat these as wellness support, not a cure.
THE STAMETS STACK
The best-known pairing: microdose + Lion's Mane (Hericium erinaceus) for nerve-growth-factor support + a small dose of niacin (B3) as a proposed distribution aid. Popularised by Paul Stamets for plasticity; rigorous human data is still limited.
FOCUS SYNERGY
L-theanine (100–200mg) takes the edge off any over-stimulation and pairs cleanly with the focus LUCID is used for. If you drink coffee, keep it modest on dosing days — LSD already raises arousal.
FOUNDATION
Omega-3 (DHA/EPA), magnesium and vitamin D form the cellular backdrop plasticity depends on. Unspectacular on their own, genuinely supportive over weeks.
